Epidemiology of Sjögren syndrome

Maxime Beydon, Sara McCoy, Yann Nguyen, Takayuki Sumida, Xavier Mariette, Raphaele Seror

Summary: Sjogren syndrome is a diverse autoimmune disorder that can occur alone or in association with other connective tissue diseases. The estimation of its prevalence and incidence is influenced by geographical and temporal factors. Extra-glandular manifestations, particularly lymphomas, are clear risk factors for mortality. Sjogren syndrome is closely associated with other connective tissue diseases and may have a mitigating effect on systemic lupus erythematosus and systemic sclerosis.

The Role of Early Treatment in the Management of Axial Spondyloarthritis: Challenges and Opportunities

Daniele Mauro, Giulio Forte, Denis Poddubnyy, Francesco Ciccia

Summary: Axial spondyloarthritis (axSpA) is a chronic inflammatory rheumatic disease that severely impacts the quality of life. Early diagnosis and intervention are crucial in mitigating long-term structural damage associated with the condition. However, current research on early pharmacological intervention remains inconclusive and further exploration is needed.

Associations of normal fasting glucose levels and of insulin resistance with degenerative rotator cuff tear

Hyung Bin Park, Ji-Yong Gwark, Jaehoon Jung

Summary: The study found that higher fasting glucose levels within the normal range are associated with an increased risk of degenerative rotator cuff tendon tear (RCT). Insulin resistance is also considered a risk factor for RCT.

A comparative analysis of suture-augmented and standard hamstring autograft single-bundle ACL reconstruction outcomes: short-term functional benefits without long-term impact

Reza Tavakoli Darestani, Sina Afzal, Ali Pourmojarab, Mojtaba Baroutkoub, Shahram Sayyadi, Hasan Barati

Summary: This study aimed to compare the outcomes of patients undergoing ACL reconstruction with or without suture augmentation. The short-term functional outcomes were better in the suture augmentation group, although there was no significant difference in complication rates between the two groups at the 24-month follow-up.

C5a stimulation induces caspase-1 activation and mature IL-1β production in human peripheral blood mononuclear cells

Yuya Fujita, Haruki Matsumoto, Kenji Inada, Michio Onizawa, Kenji Saito, Yuya Sumichika, Shuhei Yoshida, Jumpei Temmoku, Naoki Matsuoka, Tomoyuki Asano, Shuzo Sato, Takeshi Machida, Kiyoshi Migita

Summary: The complement component C5a contributes to the recruitment of immune cells and local inflammation. The activation of inflammasome and the production of proinflammatory cytokine IL-1 beta are also involved in inflammatory disorders. This study investigates the association between inflammasome activation and C5a and reveals that C5a stimulates the production of IL-1 beta mainly in monocytes through caspase-1 activation.

Wilforine inhibits rheumatoid arthritis pathology through the Wnt11/β-catenin signaling pathway axis

Yurong Huang, Yanhui Peng, Hui Li, Chen Li, Yajie Wu, Xiaomei Wang, Jun Chang, Chenggui Miao

Summary: This study reveals that WFR improves rheumatoid arthritis by inhibiting the Wnt11/beta-catenin signaling pathway. Wnt11 is a direct target of WFR, and WFR has a significant inhibitory effect on FLS proliferation.

Impact of the COVID-19 Pandemic on Health Care Access and Diagnosis of Pulmonary Arterial Hypertension Among Patients With Systemic Sclerosis

Alicia Callisto, Dylan Hansen, Jenny Walker, Gene Siew Ngian, Diane Apostolopoulos, David Liew, Vibhasha Chand, Catherine L. Hill, Kim Griggs, Alicia Calderone, Mandana Nikpour, Joanne Sahhar, Wendy Stevens, Susanna Proudman

Summary: This study aimed to investigate the impact of COVID-19 pandemic-related restrictions on health care access of patients with systemic sclerosis (SSc). The results showed that the pandemic led to worse mental health and reduced screening and diagnosis of pulmonary arterial hypertension (PAH) in SSc patients, which may have long-term implications.

Summary: This study evaluated the potential of Sema4D as a marker for rheumatoid arthritis (RA) disease activity and found that Sema4D levels effectively reflect moderate to severe RA activity. Combining Sema4D with other known biomarkers increases the diagnostic power of RA activity. The multiple regression algorithm method shows promise in calculating disease activity.

New genes, pathways and therapeutic targets in autoinflammatory diseases

Riccardo Papa, Marco Gattorno

Summary: Studies in 2023 have discovered eight new monogenic autoinflammatory diseases and the mutations causing them. These studies have provided insights into clinical phenotypes, pathogenic mechanisms and potential therapies. The research has also identified autoinflammatory pathways related to mitochondrial dysfunction or overactivation of SRC family kinases. In addition, a new autoinflammatory disease called LIRSA has been characterized, highlighting the role of IL-1 in sterile bone inflammation and leading to the development of an IL-1 inhibitor. These findings contribute to understanding the pathogenesis of these diseases and offer new treatment options.

Eric Tille, Franziska Beyer, Cornelia Luetzner, Anne Postler, Peter Thomas, Burkhard Summer, Joerg Luetzner

Summary: This study compared coated and standard total knee arthroplasty (TKA) in terms of inflammatory response and patient-reported outcome measures (PROMs). The results showed no significant differences in function and quality of life between the assessed groups at any follow-up. There were no relevant increases in serum cytokine levels, even in allergy patients. Additionally, there were no differences between coated and standard TKA in non-allergy patients during the 3-year follow-up period.

Respiratory infection risk in primary Sjögren's syndrome complicated with interstitial lung disease: a retrospective study

Xingyu Zhou, Hao Li, Naidi Wang, Yuebo Jin, Jing He

Summary: The objective of this study was to explore the clinical and laboratory characteristics of pSS-ILD and investigate the risk factors for respiratory infections. The results showed that non-specific interstitial pneumonia was the most common type of ILD, and bacteria were the most common pathogens in infected patients. Higher levels of ESSDAI, CRP, ESR, and C3 may be associated with an increased risk of infection, while PAH and reduction of DLCO were identified as independent risk factors.
