Enzalutamide and olaparib synergistically suppress castration-resistant prostate cancer progression by promoting apoptosis through inhibiting nonhomologous end joining pathway

Hui-Yu Dong, Pan Zang, Mei-Ling Bao, Tian-Ren Zhou, Chen-Bo Ni, Lei Ding, Xu-Song Zhao, Jie Li, Chao Liang

Summary: Recent studies have found that the combination of anti-androgen enzalutamide and PARP inhibitor olaparib can significantly inhibit the proliferation and induce apoptosis in AR-positive prostate cancer cells. Bioinformatics analysis revealed that the combination treatment affects nonhomologous end joining and apoptosis pathways. Moreover, enzalutamide enhances the response of prostate cancer cells to the combination therapy by regulating anti-apoptotic and pro-apoptotic genes.

Apalutamide for metastatic castration-sensitive prostate cancer: final analysis of the Asian subpopulation in the TITAN trial

Byung Ha Chung, Jian Huang, Hiroji Uemura, Young Deuk Choi, Zhang-Qun Ye, Hiroyoshi Suzuki, Taek Won Kang, Da-Lin He, Jae Young Joung, Sabine D. Brookman-May, Sharon McCarthy, Amitabha Bhaumik, Anildeep Singh, Suneel Mundle, Simon Chowdhury, Neeraj Agarwal, Ding-Wei Ye, Kim N. Chi, Hirotsugu Uemura

Summary: The final analysis of the TITAN trial demonstrated that apalutamide plus androgen deprivation therapy (ADT) improved overall survival and other efficacy endpoints in patients with metastatic castration-sensitive prostate cancer (mCSPC). A post hoc final analysis focusing on the Asian subpopulation showed consistent efficacy and safety of apalutamide in Asian patients, with reduced risks of death, castration resistance, prostate-specific antigen (PSA) progression, and second progression-free survival (PFS2) compared to placebo.

Can a sperm selection technique improve embryo ploidy?

Olena M. M. Kocur, Philip Xie, Stephanie Cheung, Sydney Souness, Mary McKnight, Zev Rosenwaks, Gianpiero D. D. Palermo

Summary: This study aimed to investigate the relationship between sperm chromatin fragmentation (SCF) and embryo aneuploidy, and to explore treatment options. The results showed that microfluidic sperm selection (MFSS) can reduce SCF and improve embryo development, resulting in increased rates of embryo euploidy and implantation.

A comparative study of two single-stage oral mucosal substitution urethroplasty (Kulkarni and Asopa) in the surgical treatments of lichen sclerosus urethral strictures

Xiang Wan, Hai-Jun Yao, Min-Kai Xie, Jian-Shu Ni, Da-Jun Gao, Zhong Wang, Bin Xu, Da-Chao Zheng

Summary: This retrospective study investigated the outcomes and complications of Kulkarni and Asopa procedures in the surgical treatment of long-segment lichen sclerosus (LS) urethral stricture. The results showed that both techniques provided acceptable results, but there was a significant difference in the recurrence-free survival rate between the two procedures. The use of antiplatelet/anticoagulant therapy, diabetes, coronary heart disease, smoking (current/former), and stricture length were identified as potential contributing factors to complications.

Effects of TAPP Combined with Biological Mesh on Reproductive Function in Male Inguinal Hernia Patients of Reproductive Age: A Single-Center Retrospective Study

Li-Xiao Zhang, Jia-Qi Kang, Zhi-Bin Ye, Li-Fei Zhang, Hong-Yu Zhao, Jiang-Nan Ge, Wei Liang

Summary: This study aimed to explore the impact of laparoscopic trans-abdominal preperitoneal hernia repair (TAPP) combined with a biological mesh on the reproductive function of male inguinal hernia patients during the reproductive period. The results showed that inguinal hernia had no significant effect on the volume and blood perfusion of the male testis. Additionally, TAPP combined with a biological mesh did not negatively affect the postoperative reproductive function of male inguinal hernia patients of childbearing age, indicating that it is a safe and feasible surgical method.

Decreased grey matter volume in dorsolateral prefrontal cortex and thalamus accompanied by compensatory increases in middle cingulate gyrus of premature ejaculation patients

Songzhan Gao, Jianhuai Chen, Jia Liu, Yichun Guan, Rusheng Liu, Jie Yang, Xianfeng Yang

Summary: This study investigated the grey matter volume (GMV) changes in the prefrontal-cingulate-thalamic areas in premature ejaculation (PE) patients and their association with ejaculatory control. The results showed impaired GMV in the dorsolateral prefrontal cortex and thalamus of PE patients, indicating their involvement in the central pathological neural mechanism underlying decreased ejaculatory control. Additionally, increased GMV in the middle cingulate gyrus suggested a compensatory mechanism for PE.

Testicular function after non-cytotoxic and immunotherapy drug treatment

David J. Handelsman, Amanda Idan, Sue Sleiman, Fey Bacha, Georgina V. Long, Alexander M. Menzies, Tejnei Vaishnav, Noosha Litkouhi, Xanthie Volckmar, William Ledger, Antoinette Anazodo

Summary: The effects of non-cytotoxic and immunotherapy drugs on human testicular function in cancer patients have not been systematically studied. This study aimed to investigate the effects of these drugs in cancer patients who had not undergone gonadotoxic treatment. The results showed that non-cytotoxic and immunotherapy drugs moderately affected testicular exocrine function (sperm production) and its hormonal correlates (follicle-stimulating hormone, inhibin B), but had minimal impact on testicular endocrine function (testosterone, luteinizing hormone).

Male obesity: Associated effects on fertility and the outcomes of offspring

Greeshma Venigalla, Vishal Ila, James Dornbush, Ari Bernstein, Justin Loloi, Edoardo Pozzi, David Miller, Ranjith Ramasamy

Summary: Male obesity has negative effects on fertility and offspring outcomes, including changes in semen quality, difficulty with natural conception, and worsened assisted reproductive technology outcomes. These effects may be modifiable with weight loss. Paternal obesity may also influence fetal development and placental abnormalities, which may be moderated by epigenetic pathways. More research is needed to understand the complex relationships involved.

The role of red ginseng in men's reproductive health: a literature review

Hao Wang, Jiwei Zhang, Dongyue Ma, Ziwei Zhao, Bin Yan, Fu Wang

Summary: Red ginseng (RG), a traditional herb commonly used in China, Korea, and other East Asian countries, has demonstrated a better clinical value in men's reproductive health (MRH). This review examines the effects of RG treatment on MRH, including its potential application in male infertility, erectile dysfunction, and prostate diseases. The mechanisms of action of RG in MRH may involve oxidative stress, regulation of sex hormones and spermatogenesis-related proteins, and anti-inflammation.
