Analysis of Low Abundance Diferential Proteins in Plasma from Hepatocellular Carcinomas Patients by Multi-dimensional Chromatography Tandem Mass Spectrometry

A multi-dimensional separation techniques was developed to analyze differential plasma proteins of immunodepleted plasma between five cases of patients with hepatocellular carcinoma (HCC) for the study and five cases of healthy volunteers for the control of plasma mixed. Samples of human plasma were depleted for most abundance proteins, using an immunoaffinity chromatography Agilent MARS Spin Cartride, then were separated under the preffered conditions by reversed phase LC. Over-expressed differential fraction of low abundance proteins were collected and trypsinized, and then the peptides trypsinzied were enriched and fractioned and indentified, using HPLC-CHIP/MS. Reverse-phase high-performance liquid chromatographic prefraction of immunodepleted human plasma coupled with HPLC-CHIP/MS to enhance mass spectrometric identification of low-abandance proteins and about 27 kinds of different protein were identified. 23 kinds of protein are related to various diseases or tumors, among them, insulin-like growth factor 2 mribonucleic acid-binding protein 3 (IMP3), aryl hydrocarbon receptor nuclear translocator (ARNT2) and glutamate receptor-interacting protein 1 (GRIP1) may become HCC diagnosis of potential biomarkers.
