Relative expression of AKR1C family members and 5 alpha R1 were determined using gene specific quantitative real-time PCR method. Both ARK1C1 and AKR1C2 were found significantly reduced in ovarian tumor tissues compared to ovarian normal tissues. In contrast, AKR1C3 and 5aR1 were kept the same transcriptional levels between ovarian tumor tissues and ovarian normal tissues. Immunohistochemical staining indicates that AKR1C1 shares the same localization with AKR1C3. Loss of ARK1C1 and AKR1C2 in ovarian cancerous tissues may enhance progesterone signaling in ovary itself by decreasing the ability of progesterone metabolism. (C) 2005 Elsevier Ltd.
