Nanozyme with the dual identity of enzymes and nanomaterials is an emerging strategy to mimic natural enzymes in bio-nanotechnology. However, how to improve the nanozyme-based therapeutic efficiency in vivo still remains a challenge. Herein, we fabricated a nanozyme platform (Fe@Fe3O4@Cu2-xS, named as MNPs) possessing peroxidase enzyme-like activity and then introduced beta-lapachone (La) to assemble an effective nanozyme (LaMNPs) to boost the production of reactive oxygen species and upregulate the level of H2O2 by the released La in an acidic tumor microenvironment. As a proof of concept, LaMNPs were injected intravenously into mice model, resulting in a significant and efficient inhibition of the tumor growth.
