Organs-on-chips: into the next decade

Summary: OoCs, also known as microphysiological systems or 'tissue chips', have the potential to be informative at multiple stages of drug discovery and development, providing insights into normal human organ function and disease pathophysiology, as well as predicting the safety and efficacy of investigational drugs in humans. The field of OoCs has seen significant advances in recent years, but there are still challenges and opportunities that need to be addressed to fully realize their potential for translational research.

Infiltration of immune cells into CRC tumors accurately predicts disease progression and patient survival. Preclinical model systems are crucial for studying immunity in the CRC TME and the utility of immunotherapies for CRC patients, with various in vitro and in vivo models available for research.

Summary: Solid tumors create a suppressive environment that impairs the cytotoxic capacity of NK cells, leading to compromised surveillance and tolerance of tumors. NK cell exhaustion persists for a period of time after removal, but can be alleviated by the addition of checkpoint inhibitors and immunomodulatory agents.
