Diterpenoids isolated from the root of Salvia miltiorrhiza and their anti-inflammatory activity

Summary: Four new diterpene-type compounds and twelve known compounds were isolated from the roots of Salvia miltiorrhiza, and their ability to inhibit nitric oxide production was evaluated. Compound 11 showed a strong inhibitory effect with an IC50 value of 3.4 +/- 1.2 μM.

Multiplexed CRISPR/Cas9-Mediated Knockout of Laccase Genes in Salvia miltiorrhiza Revealed Their Roles in Growth, Development, and Metabolism

Summary: In this study, the CRISPR/Cas9 system was used to target conserved domains for knockout of multiple genes in the laccase family in S. miltiorrhiza. The gene-edited lines showed significantly decreased expression of target laccase genes and key genes involved in phenolic acid biosynthesis. The growth and development of hairy roots in the edited lines were significantly retarded, with abnormal root development and larger, looser xylem cells observed. Additionally, the accumulation of key compounds like salvianolic acid B was decreased, indicating the essential roles of SmLACs in root development and lignin formation in S. miltiorrhiza.

Summary: The endophytic fungus Mucor circinelloides DF20 can significantly promote the biosynthesis and accumulation of tanshinones in the root of Salvia miltiorrhiza, especially tanshinone IIA content, which is 22 times that of the control group. DF20 hyphae mainly colonize in the root tissue interspace of S. miltiorrhiza, with some located inside the cells. DF20 colonization can increase the expression level of key enzyme genes in the tanshinone biosynthesis pathway, mainly in the early stage of co-culture.
