Hypoxia-degradable zwitterionic phosphorylcholine drug nanogel for enhanced drug delivery to glioblastoma

Summary: In this study, a hypoxia-degradable zwitterionic phosphorylcholine nanogels (PMPC)-P-H was developed for enhanced chemotherapy of glioblastoma. The nanogels exhibited favorable biocompatibility, long-circulating property, and the ability to pass through the blood-brain barrier effectively, leading to long-lasting accumulation in glioblastoma tissue and fast drug release in hypoxic environment. Overall, the (PMPC)-P-H drug nanogels showed promising tumor inhibition effect in a glioblastoma model, with negligible side effects, offering a potential nanoplatform for treating hypoxic relevant diseases in the central nervous system.

Ultra-small size gelatin nanogel as a blood brain barrier impermeable contrast agent for magnetic resonance imaging

Summary: In this study, ultra-small gelatin nanogels were developed as blood-brain-barrier impermeable contrast agents for MRI. Using a radiation crosslinking technique, the integrity of amino acids in the gelatin nanogels was ensured while conjugating with gadolinium to form GdGN. The safety and efficacy of GdGN as an MRI contrast agent were verified through in vivo studies, showing rapid renal excretion within 90 minutes and no passage across brain barriers.

Low nanogel stiffness favors nanogel transcytosis across an in vitro blood-brain barrier

Summary: This study investigated the effect of nanoparticle stiffness on transport across the blood-brain barrier, finding that more densely cross-linked nanogels showed higher uptake by brain endothelial cells, while less densely cross-linked nanogels demonstrated higher transcytotic potential. These findings suggest that nanogel stiffness has opposing effects on nanogel uptake and transcytosis at the BBB.

It estimated that there were nearly 19.3 million new cancer cases and almost 10 million cancer deaths worldwide in 2020, with the most commonly diagnosed cancers being female breast cancer and lung cancer, and the most common causes of cancer death being lung cancer and liver cancer.
