Delta-24-RGD, an Oncolytic Adenovirus, Increases Survival and Promotes Proinflammatory Immune Landscape Remodeling in Models of AT/RT and CNS-PNET

Marc Garcia-Moure et al.

Summary: The study demonstrated that Delta-24-RGD efficiently infected and replicated in various AT/RT and CNS-PNET cell lines, exhibiting significant cytotoxicity against tumor cells. In vivo, intratumoral injection of Delta-24-RGD significantly extended survival in mouse models of AT/RT and PNET, leading to long-term survival outcomes. Additionally, the viral therapy showed promising results in hindering tumor dissemination and increasing survival rates in immunocompetent humanized mouse models.

Delta-24 adenoviral therapy for glioblastoma: evolution from the bench to bedside and future considerations

Chibawanye I. Ene et al.

Summary: Delta-24-based oncolytic viruses have evolved over the years with structural improvements to enhance infectivity for cancer cells while minimizing toxicity to normal cells. Clinical trials have demonstrated potential antitumor responses with immune cell infiltration in the tumor microenvironment, suggesting a promising long-term antitumor immune response.ongoing research aims to address the remaining obstacles limiting the efficacy of Delta-24 adenovirus therapy for glioblastoma.
