How (5′S) and (5′R) 5′,8-Cyclo-2′-Deoxypurines Affect Base Excision Repair of Clustered DNA Damage in Nuclear Extracts of xrs5 Cells? A Biochemical Study

Karolina Boguszewska et al.

Summary: CDLs are a characteristic feature of ionizing radiation's impact on human genetic material, impairing cellular repair machinery efficiency, especially BER. CdPus affect nuclear BER efficiency, where RcdA inhibits BER more than ScdA, and ScdG decreases repair level more than RcdG. CdPus influence repair of the other lesion within the CDL, potentially leading to prolonged unrepaired lesions and impaired cellular processes.

The Influence of 5′R and 5′S cdA and cdG on the Activity of BsmAI and SspI Restriction Enzymes

Michal Szewczuk et al.

Summary: Restriction endonucleases (REs) are bacterial scissors that combat foreign genetic material. SspI and BsmAI are type II REs examined in this study, cleaving dsDNA at recognition sites. Ionizing radiation can damage cellular mechanisms, with 5',8-cyclo-2'-deoxypurines (cdPus) impacting DNA repair pathways. The study found that cdPus affect RE activity, with SspI inhibited by lesions at the recognition site and BsmAI activity lowered by ScdA and cdG diastereomers.
