Regio- and Enantioselective (3+3) Cycloaddition of Nitrones with 2-Indolylmethanols Enabled by Cooperative Organocatalysis

Summary: The regio- and enantioselective (3+3) cycloaddition of nitrones with 2-indolylmethanols was successfully achieved through the cooperative catalysis of hexafluoroisopropanol (HFIP) and chiral phosphoric acid (CPA). This approach resulted in the synthesis of indole-fused six-membered heterocycles with high yields, excellent enantioselectivities, and exclusive regiospecificity. The role of the cocatalyst HFIP in helping CPA control the reactivity and enantioselectivity of the reaction was elucidated through theoretical calculations, demonstrating a new mode of cooperative catalysis.

Summary: This work presents a catalytic enantioselective construction of axially chiral alkene-indole frameworks using alpha-amido sulfones as nucleophiles and chiral phosphoric acid as catalyst. The strategy resulted in high yields, excellent (E/Z)-selectivity, and good enantioselectivities, representing the first of its kind in the construction of axially chiral alkene-indole scaffolds.

Regio- and enantioselective ring-opening reaction of vinylcyclopropanes with indoles under cooperative catalysis

Summary: The study established a reaction under the cooperative bimetallic catalysis of iridium and copper catalysts, which yielded indole C3-allylation products with moderate yields (up to 78%) and good enantioselectivities (up to 97:3 er). This reaction not only represents the first catalytic asymmetric ring-opening reaction of vinylcyclopropanes with C3-unsubstituted indoles, but also provides an atom-economic and straightforward method for the synthesis of C3-allylic indoles with high regio- and enantioselectivity.
