Handhold-Mediated Strand Displacement: A Nucleic Acid Based Mechanism for Generating Far-from-Equilibrium Assemblies through Templated Reactions

Javier Cabello-Garcia et al.

Summary: The use of templates for sequence-controlled assemblies, particularly in long polymers, is a well-established method. However, the transient template recognition interactions during the biogenesis of sequence-controlled polymers can disrupt the equilibrium, leading to the production of far-from-equilibrium output. Handhold-mediated strand displacement (HMSD) is proposed as an alternative mechanism to generate out-of-equilibrium products by decoupling drive and specificity in DNA-based reactions.

Coacervates as models of membraneless organelles

N. Amy Yewdall et al.

Summary: Coacervates are condensed liquid-like droplets formed by oppositely charged polymeric molecules, with remarkable material properties that have been extensively studied. They play an important role in the formation of membraneless organelles in living cells. Well-characterized coacervates can serve as useful models to better understand specific aspects of membraneless organelles.

Self-assembly of aramid amphiphiles into ultra-stable nanoribbons and aligned nanoribbon threads

Ty Christoff-Tempesta et al.

Summary: The study introduces a small-molecule platform called aramid amphiphile, which exhibits stability and can self-assemble in water to form nanoribbons with high strength and toughness. These nanoribbons can be further organized into dry threads that can support 200 times their weight, extending small-molecule self-assembly to macroscopic materials and enabling new possibilities for solid-state applications.

DNA origami signposts for identifying proteins on cell membranes by electron cryotomography

Emma Silvester et al.

Summary: Electron cryotomography (cryoET) has revolutionized our understanding of biological function by revealing molecular details of membranes, viruses, and cells. A new tagging strategy using DNA origami allows for precise identification of individual protein complexes in tomograms without relying on metal clusters, making it suitable for a wide range of biological surfaces in cryoET studies.

Reversible membrane deformations by straight DNA origami filaments

Henri G. Franquelim et al.

Summary: The study demonstrates the formation and effects of membrane-bound filaments using DNA origami to mimic cytoskeletal elements. By manipulating the MgCl2 concentration, control over the polymerization and depolymerization of DNA origami is achieved, leading to the formation of helical structures bound to the membrane. This approach provides a new insight into shaping membranes by straight cytoskeletal-like objects.
