The Development of Hsp90β-Selective Inhibitors to Overcome Detriments Associated with pan-Hsp90 Inhibition

Sanket J. Mishra et al.

Summary: Hsp90 proteins play a crucial role in cancer progression, and inhibiting their activity may be beneficial for cancer treatment. Current Hsp90 inhibitors target all isoforms, potentially leading to adverse effects. Utilizing Hsp90 beta-selective inhibitors as a new approach for cancer therapy holds promise in reducing side effects.

Visualizing the Dynamics of a Protein Folding Machinery: The Mechanism of Asymmetric ATP Processing in Hsp90 and its Implications for Client Remodelling

Ilda D'Annessa et al.

Summary: The Hsp90 chaperone system forms dynamic multiprotein complexes with a wide range of client proteins, involving cochaperone partners to regulate client activities, protect them from degradation, and respond to biochemical signals. Cryo-electron microscopy has provided a molecular picture of Hsp90 in complex with a kinase and a cochaperone. Structural modulations dependent on nucleotides lead to asymmetry in Hsp90 dynamics, revealing specific long-range coordination patterns between nucleotide binding, client binding, cochaperone, and client regions.
