It has been demonstrated that catenated single-stranded DNA circles can be used as a universal scaffold for the production of topologically linked DNA origami structures, with assembly achieved via a simple one-pot annealing protocol.
DNA origami is awidely used DNA nanotechnology that allows constructionof two-dimensional and three-dimensional nanometric shapes. The designabilityand rigidity of DNA origami make it an ideal material for constructionof topologically linked molecules such as catenanes, which are attractivefor their potential as motors and molecular machines. However, a generalmethod for production of topologically linked DNA origami has beenlacking. Here, we show that catenated single-stranded DNA circlescan be produced and used as a universal scaffold for the productionof topologically linked (catenated) DNA origami structures where theindividual linked structures can be of any arbitrary design. Assemblyof these topologically linked DNA origami structures is achieved viaa simple one-pot annealing protocol.
