Chelating Phosphine-N-Heterocyclic Carbene Platinum Complexes via Catalytic Asymmetric Hydrophosphination and Their Cytotoxicity Toward MKN74 and MCF7 Cancer Cell Lines

Jeffery Wee Kiong Seah et al.

Summary: In this study, a series of activated vinyl azoles were hydrophosphinated under mild conditions in the presence of a chiral palladacycle catalyst to give enantioenriched phosphine azoles with moderate enantioselectivities and yields. The racemic phosphine azoles were further transformed into novel chelating phosphine-N-heterocyclic carbene (NHC) platinum complexes, and the drug efficacies of nine selected compounds were evaluated in two cancer cell lines, with two compounds showing activities comparable to that of cisplatin.

Access to a Chiral Phosphine-NHC Palladium(II) Complex via the Asymmetric Hydrophosphination of Achiral Vinyl Azoles

Jeffery Wee Kiong Seah et al.

Summary: Enantioenriched phosphine azoles were successfully synthesized via palladium-catalyzed asymmetric hydrophosphination with excellent yields and enantioselectivities under mild reaction conditions. One of the phosphine azoles was methylated and complexed to palladium(II) cleanly, resulting in a chiral phosphine-NHC palladium(II) complex with excellent overall conversion.
