Chromatin mechanics dictates subdiffusion and coarsening dynamics of embedded condensates

Daniel S. W. Lee et al.

Summary: The study demonstrates that droplet growth dynamics are directly inhibited by the chromatin-dense environment, leading to an anomalously slow coarsening exponent. Through scaling arguments and simulations, the researchers show that this arrested growth can arise due to subdiffusion of individual condensates. Tracking the fluctuating motion of condensates within chromatin reveals a subdiffusive exponent, explaining the anomalous coarsening behaviour.

Self-Assembly of Biomolecular Condensates with Shared Components

William M. Jacobs

Summary: Biomolecular condensates are droplets formed by proteins and nucleic acids spontaneously separating in the crowded intracellular environment, serving as the foundation for membraneless compartments in cells. Research shows that in a system that may not be at thermodynamic equilibrium, the number of reliably assembled distinct condensates increases superlinearly with the number of species when they share components. Additionally, it is possible to predict the maximum number of distinct condensates in a mixture without knowing the details of pairwise interactions.

Mechanical Frustration of Phase Separation in the Cell Nucleus by Chromatin

Yaojun Zhang et al.

Summary: The mechanical properties of chromatin were found to suppress coalescence and ripening of liquid droplets in the nucleus, quantitatively controlling their number and size. The study highlights the importance of the subcellular mechanical environment in regulating condensates.

Regulation of biomolecular condensates by interfacial protein clusters

Andrew W. Folkmann et al.

Summary: This study reveals that the dynamics of biomolecular condensates are regulated by protein clusters. Through experiments and theory, it is demonstrated that the assembly of P granules is controlled by MEG-3 protein, which helps lower surface tension and slow down coarsening.
