Summary: This study focuses on the activation mechanisms of PI3Kα, particularly driven by double activating mutations. It highlights the importance of strong driver mutations in fully activating PI3K, as single strong drivers may not be sufficient. In contrast, double strong drivers can effectively activate PI3K, and interactions between different driver mutations play a critical role in the activation process.

Summary: circ-PRMT5 is up-regulated in breast cancer tissues and cells, and its high expression is associated with poor prognosis in patients. It suppresses cell proliferation and angiogenesis, and promotes cell apoptosis. Mechanistically, circ-PRMT5 acts as a miR-509-3p sponge to up-regulate TCF7L2 expression. PKBβ/AKT2 deficiency impacts brain mTOR signaling, prefrontal cortical physiology, hippocampal plasticity and select murine behaviors

Sara Palumbo et al.

Summary: The serine/threonine protein kinase v-AKT homologs (AKTs) play important roles in neurodevelopment, with Akt2 specifically involved in axonal development, dendritic patterning, and neural circuit physiology. Akt2 loss-of-function leads to impairments in anxiety, fear conditioned learning, social behaviors, and memory, as well as dysregulated mTOR and GSK3 beta signaling in the brain. Pharmacological enhancement of PI3K/AKT signaling and PIK3CD inhibition can rescue cognitive impairments associated with Akt2 deficiency.
