Summary: The study demonstrated that beta GalCer is an optimized lipid anchor, enabling the noncovalent vaccine candidate GM3-beta GalCer/alpha GalCer to evoke a comparable antibody level to GM3-alpha GalCer. However, the antibodies induced by GM3-alpha GalCer show better recognition of B16F10 cancer cells and more efficiently activate the complement system.

Using agonists for iNKT cells in cancer therapy

Summary: α-Galactosylceramide (α-GalCer) shows potential as an anti-cancer agent by stimulating iNKT cells, but low frequencies and hypo-responsiveness in cancer patients have limited its efficacy. Current clinical applications focus on expanding iNKT cells ex vivo for adoptive therapy, and strategies are being explored to improve in vivo presentation and enhance APC-licencing function for adaptive immune responses. A new observation suggests in vivo activated iNKT cells may play a role in forming resident memory CD8(+) T cells for cancer therapy in the future.

Full synthesis and bioactivity evaluation of Tn-RC-529 derivative conjugates as self-adjuvanting cancer vaccines

Summary: An efficient strategy for constructing RC-529 and its derivatives has been established, with four conjugates of RC-529 derivatives with Tn antigen inducing strong T cell dependent immune responses in mice and showing specific cytotoxicity towards Tn-overexpressing cancer cells. This suggests RC-529 as a potential platform for the development of new vaccine carriers with self-adjuvanting properties for cancer treatment, supported by preliminary structure-activity relationship analysis.
