Synthesis and Evaluation of Arylamides with Hydrophobic Side Chains for Insulin Aggregation Inhibition

Summary: By synthesizing a small library of oligopyridylamide foldamers decorated with different hydrophobic side chains, a hit molecule BPAD2 was identified with an IC50 value of 0.9 μM for inhibiting insulin aggregation. Mechanistic analyses showed that BPAD2 mainly inhibited secondary nucleation and elongation processes during aggregation, highlighting the importance of structure-activity relationship studies in understanding protein aggregation at a molecular level.

The Effect of Cholesterol on Membrane-Bound Islet Amyloid Polypeptide

Summary: This study used atomistic, unbiased molecular dynamics simulations to investigate the effect of cholesterol on membrane bound IAPP. Results indicated that cholesterol decreased the insertion depth of IAPP compared to pure phospholipid membranes, while phosphatidylserine (PS) lipids counteracted the effect of cholesterol. The depletion of cholesterol in the vicinity of IAPP suggests that its effect is indirect.

Alizarin and Purpurin from Rubia tinctorum L. Suppress Insulin Fibrillation and Reduce the Amyloid-Induced Cytotoxicity

Summary: Natural anthraquinone compounds alizarin and purpurin have been found to inhibit insulin fibrils formation and reduce insulin-induced cytotoxicity, with purpurin showing a more significant inhibitory effect. Computer simulations suggested that these compounds interact mainly with hydrophobic residues of insulin chain B and interfere with phenylalanine residue binding. This research indicates the potential of natural anthraquinone compounds in preventing protein misfolding diseases and designing effective antiamyloidosis compounds.

Prion-derived tetrapeptide stabilizes thermolabile insulin via conformational trapping

Summary: A new tetrapeptide VYYR has been found to effectively inhibit insulin fibrillation and maintain its bioactivity, providing good blood sugar control in mouse models of diabetes. The critical interaction between insulin and IS1 has been elucidated through nuclear magnetic resonance spectroscopy, and a non-canonical dimer-mediated conformational trapping mechanism has been demonstrated for insulin stabilization. This study introduces a class of tetrapeptides for developing thermostable insulin formulations with therapeutic relevance.
