Summary: Metabolomics is an important field with challenges in biomarker qualification for clinical applications, due to issues such as inappropriate study designs and insufficient data to support clinical utility. To address these challenges, modifications of existing guidelines and alternative approaches are being developed to facilitate method validation. The main challenges in method development and validation for endogenous metabolites include matrix effects evaluation, alternative analyte-free matrices, and the choice of internal standards.

Summary: This study aimed to establish a robust method for dual visualization of miR and protein by combining chromogen-based in situ hybridization and immunohistochemistry, and apply it to investigate stromal expression of miR204 as a putative prognostic biomarker in oral squamous cell carcinoma. The study found that miR204 stromal expression at the tumor front predicted recurrence-free survival (p = 0.032) and overall survival (p = 0.036), and was further confirmed as an independent prognostic biomarker in OSCC through multivariate Cox regression analysis.
