Interleukin-6 produced by enteric neurons regulates the number and phenotype of microbe-responsive regulatory T cells in the gut

Yiqing Yan et al.

Summary: The immune and enteric nervous systems interact with each other, with microbial signals potentially influencing neuronal density and activation, thereby regulating the generation of Treg cells and immunological tolerance in the gut.

Disorders of the enteric nervous system - a holistic view

Beate Niesler et al.

Summary: The enteric nervous system (ENS) plays a crucial role in congenital enteric neuropathic disorders, but its role in systemic diseases is less understood. Evidence suggests that the ENS is emerging as a key player in neurodegenerative diseases, cancer, and diabetes, regulating gut function and contributing to various disorders by interacting with the gut microbiome, genetic background, central nervous system, and immune system.

A Selective α7 Nicotinic Acetylcholine Receptor Agonist, PNU-282987, Attenuates ILC2s Activation and Alternaria-Induced Airway Inflammation

Fang Yuan et al.

Summary: The study found that PNU-282987 significantly reduced goblet cell hyperplasia, eosinophil infiltration, and ILC2 numbers in the airways following IL-33 or AA challenges. In vitro experiments showed a decrease in GATA3 and Ki67 expression in isolated lung ILC2s after treatment with PNU-282987 or GTS-21. Additionally, PNU-282987 treatment resulted in decreased IKK and NF-kappa B phosphorylation compared to GTS-21 in ILC2s.

Acetylcholine production by group 2 innate lymphoid cells promotes mucosal immunity to helminths

Luke B. Roberts et al.

Summary: ILC2s are shown to synthesize and release acetylcholine during parasitic nematode infection. Exposure to extracts of certain substances or factors in vivo can promote the synthesis of acetylcholine by ILC2s. The ACh derived from ILC2s plays a functional role in expanding ILC2s for maximal induction of type 2 immunity.

The ChAT-acetylcholine pathway promotes group 2 innate lymphoid cell responses and anti-helminth immunity

Coco Chu et al.

Summary: ILC2s up-regulate ChAT and express AChRs after infection or treatment, with ACh administration promoting cytokine production and helminth expulsion. Deficiency of ChAT in ILC2s leads to impaired immunity.

Optogenetic activation of local colonic sympathetic innervations attenuates colitis by limiting immune cell extravasation

Maya Schiller et al.

Summary: This study investigated the impact of the local arm of the sympathetic nervous system on colonic inflammation. It was found that local activation of sympathetic fibers can attenuate colitis and reduce immune cell abundance. Gene expression profiling showed that optogenetic stimulation leads to decreased endothelial expression of the adhesion molecule MAdCAM-1, which can be blocked by adrenergic blockers and 6-hydroxydopamine. Antibody blockade of MAdCAM-1 eliminated the effect of optogenetic stimulation on immune cell extravasation and pathology in the colon.
