Summary: The study established a multifunctional drug delivery nanosystem (HTLAC) for rapid response to the DTD enzyme, leading to precise detection and termination of tumors. By incorporating DTD-responsive substrates and encapsulating drugs in cell-penetrating peptide and hyaluronic acid functionalized liposomes, HTLAC achieved enhanced tumor detection and superior antitumor efficiency. The HTLAC nanosystem provides an efficacious approach for the fabrication of enzyme-triggering nanosystems to detect tumors and induce the self-burst of ROS for efficient tumor treatment.
