Discovery of Benzocyclic Sulfone Derivatives as Potent CXCR2 Antagonists for Cancer Immunotherapy

Summary: The study designed benzocyclic sulfone derivatives as potent CXCR2 antagonists, with lead compounds 9b and 11h showing high inhibitory effects on tumor growth in the Pan02 mouse model through oral administration. They also reduced neutrophils and MDSCs infiltration while enhancing CD3(+) T lymphocytes infiltration into tumor tissues.

Discovery of 1,5-Dihydro-4H-imidazol-4-one Derivatives as Potent, Selective Antagonists of CXC Chemokine Receptor 2

Summary: The study demonstrated the critical roles of CXCR1 and CXCR2 in cancer metastasis, but designing selective antagonists for these receptors remains unclear. Using a pharmacophore model, a selective CXCR2 antagonist compound was identified, and further optimization led to compound C5 with higher selectivity. Dynamic simulation revealed the key amino acid residue contributing to the selectivity of C5, providing important clues for designing new CXCR2 selective antagonists.

Anticancer opportunities at every stage of chemokine function

Summary: The chemokine system, with 48 chemokines and 23 receptors, plays a critical role in cancer and various pharmacological approaches have been developed to intervene at different stages of chemokine function. Despite efforts from the pharmaceutical sector, only two drugs are currently approved for clinical use against cancer. Many of the strategies have been tested in preclinical cancer models, with some advancing to clinical trials as potential anticancer therapies.
