Overproduction of α-Farnesene in Saccharomyces cerevisiae by Farnesene Synthase Screening and Metabolic Engineering

Summary: This study aimed to maximize the flux of farnesyl diphosphate to farnesene biosynthesis in Saccharomyces cerevisiae. Through screening and genetic engineering, a high alpha-farnesene producing yeast strain was developed, reaching a production level of 10.4 g/L. Various strategies were employed, such as overexpressing key enzymes and optimizing gene expression, to achieve the significant improvement in farnesene production.

Yeast-Based Biosynthesis of Natural Products From Xylose

Summary: Xylose is the second most abundant sugar in lignocellulosic hydrolysates, and utilizing yeast strains for transforming xylose into valuable chemicals is a feasible and sustainable approach. Metabolically engineered yeasts have shown significant progress in producing natural products from xylose, such as aromatics, terpenoids, and flavonoids. Challenges and future perspectives in yeast engineering for commercial production of natural products using xylose as feedstocks are discussed.

Methanotrophic microbial cell factory platform for simultaneous conversion of methane and xylose to value-added chemicals

Enhancing Sesquiterpenoid Production from Methane via Synergy of the Methylerythritol Phosphate Pathway and a Short-Cut Route to 1-Deoxy-D-xylulose 5-Phosphate in Methanotrophic Bacteria

Summary: This study demonstrated the use of a shortcut route from ribulose 5-phosphate to DXP for more efficient sesquiterpenoid production, with a significant increase achieved by the fusion of nDXP enzyme and DXP reductase. Engineering strategies were also applied to produce another sesquiterpenoid, alpha-bisabolene, showing a twofold improvement in titer compared to the initial strain. The synergistic use of the nDXP and MEP pathways in methanotrophic bacteria was shown to enhance sesquiterpenoid production.
