Increased circulating CRTH2(+)Tregs are associated with asthma control and exacerbation

Teerapol Chantveerawong et al.

Summary: The frequency of CRTH2(+)Tregs is higher in patients with asthma, especially allergic phenotype, compared to healthy controls, and shows a negative correlation with ACT scores and PEFR. CRTH2(+)Tregs increase during exacerbation and decrease after asthma control. In vitro investigations demonstrate that Tregs of allergic patients with asthma display Th2-biased character.

Bilirubin represents a negative regulator of ILC2 in allergic airway inflammation

Juan He et al.

Summary: Bilirubin, an end product of heme catabolism, was shown to be a potent negative regulator of ILC2s, suppressing their responses to IL-33 and alleviating airway inflammation. Mechanistic studies revealed that bilirubin exerts its effects on ILC2s through downregulation of ERK phosphorylation and GATA3 expression. Clinical evidence suggested that newborns with hyperbilirubinemia exhibited lower levels of functional ILC2 and suppressed ERK signaling. These findings suggest that bilirubin may have therapeutic potential in the treatment of allergic airway inflammation.

Neuropilin-1 mediates lung tissue-specific control of ILC2 function in type 2 immunity

Jingjing Zhang et al.

Summary: This study identifies neuropilin-1 (Nrp1) as a tissue-specific marker of lung-resident ILC2s, which enhances their function and type 2 immune response by upregulating IL-33 receptor ST2 expression. Inhibition of Nrp1 suppresses IL-5 and IL-13 production by ILC2s and protects mice from the development of pulmonary fibrosis.

Summary: This study investigates the phenotypic and molecular features of cutaneous innate lymphoid cells (ILCs) to better understand the biological role of these cells. The findings demonstrate the flexibility of the skin immune system, with ILCs in diseased skin expressing genes characteristic of different immune types. This suggests that the skin immune system can adjust its function to adapt to the environment.

IL-10 production by ILC2s requires Blimp-1 and cMaf, modulates cellular metabolism, and ameliorates airway hyperreactivity

Emily Howard et al.

Summary: The study identified a unique subset of ILC2s, known as ILC2(10)s, which actively produce IL-10 to suppress proinflammatory ILC2 functions through autocrine and paracrine signaling, resulting in reduced lung inflammation and airway hyperresponsiveness in vivo. Furthermore, the research highlights the metabolic dependency of ILC2(10)s on the glycolytic pathway for IL-10 production, contrasting with the conventional fatty acid oxidation pathway used for proinflammatory effector functions.

Summary: This study utilized single-cell RNA sequencing to uncover tissue-specific transcriptional profiles and heterogeneity of CD127(+) ILCs in four human tissues. It identified migratory properties, tissue-residency, activation, and modified metabolism characteristics of ILCs in different tissues. The study also revealed potential differentiation pathways from naive to mature ILC subsets, and unique TCR-V(D)J-rearrangement patterns of blood ILC1-like cells.

Induction of IL-10-producing type 2 innate lymphoid cells by allergen immunotherapy is associated with clinical response

Korneliusz Golebski et al.

Summary: IL-10(+) ILC2s play a crucial role in modulating grass-pollen allergy, attenuating Th responses, and maintaining epithelial cell integrity. Patients with grass-pollen allergy have lower levels of IL-10(+) KLRG1(+) ILC2s, but this ability to produce IL-10 is restored after immunotherapy treatment. The mechanisms underlying this involve modifications in retinol metabolic pathway, cytokine-cytokine receptor interaction, and JAK-STAT signaling pathways in the ILCs.

Homeostatic IL-13 in healthy skin directs dendritic cell differentiation to promote TH2 and inhibit TH17 cell polarization

Johannes U. Mayer et al.

Summary: The study demonstrates that IL-13 secreted by dermal innate lymphoid cells plays a crucial role in promoting the differentiation of a CD11b(lo) type 2 dendritic cell subset in the skin of mice and humans, supporting the development of T(H)2 cells and inhibiting T(H)17 cells. This homeostatic IL-13 fosters a noninflammatory skin environment conducive to allergic sensitization.
