High-accuracy long-read amplicon sequences using unique molecular identifiers with Nanopore or PacBio sequencing

Soren M. Karst et al.

Summary: High-throughput amplicon sequencing of large genomic regions remains challenging for short-read technologies. Here, a new approach combining unique molecular identifiers (UMIs) with Oxford Nanopore Technologies (ONT) or Pacific Biosciences circular consensus sequencing was reported, achieving high-accuracy single-molecule consensus sequences. The method was successfully applied to sequence ribosomal RNA operon amplicons and genomic sequences of microbial communities, showing a very low chimera rate.

Molecular Epidemiology of Enterovirus in Children with Central Nervous System Infections

Lamprini Posnakoglou et al.

Summary: Limited recent molecular epidemiology data are available for pediatric Central Nervous System (CNS) infections in Europe. This study investigated the molecular epidemiology of enterovirus (EV) involved in CNS infections in children, finding a seasonal distribution of EV CNS infections with predominant genotypes including CV-B5, E30, E16, and E11, showing significant phylogenetic differences from previously described isolates.

Ivica Letunic et al.

Summary: The Interactive Tree Of Life is an online tool for displaying, manipulating, and annotating phylogenetic and other trees. The latest version 5 introduces a new tree display engine and several new features, such as a new dataset type and expanded annotation options. Users can now also manually annotate trees by hand, providing more flexibility and control.

Genetic diversity and evolution of enterovirus A71 subgenogroup C1 from children with hand, foot, and mouth disease in Thailand

Jiratchaya Puenpa et al.

Summary: Eleven full-length genome sequences of 2019 EV-A71 C1 strains isolated from HFMD patients in Thailand from 2019 to early 2020 were described, showing the emergence of multiple lineages through recombination. Bayesian-based phylogenetic analysis indicated that the 2019 EV-A71 C1 Thai strains share a common ancestor with variants in Europe. The substitution rate for the 2019 EV-A71 C1 genome was estimated to be similar to that observed in previous outbreaks. These findings are crucial for understanding the evolution of EV-A C1 during the ongoing HFMD outbreak and its relevance to disease outcomes in children worldwide.
