Machine Learning-Assisted Sampling of SERS Substrates Improves Data Collection Efficiency

Tatu Rojalin et al.

Summary: SERS is a powerful technique for sensitive label-free analysis, but automation progress is limited by manual sample preparation. Current automation efforts focus on downstream processing of collected data, but there is a need to develop algorithms to simplify and accelerate data collection. Machine learning-assisted methods at the acquisition stage can serve as a useful building block for point-of-care SERS diagnostic platforms.

Live Cell Poration by Au Nanostars to Probe Intracellular Molecular Composition with SERS

Evelina I. Nikelshparg et al.

Summary: A new type of flat substrate utilizing stabilized aggregates of gold nanostars and a polymer layer has been used for visualizing structures and studying biochemical processes within living cells via surface-enhanced Raman scattering (SERS). The long tips of the gold nanostars can penetrate the cell membrane to receive the SERS signal from biomolecules inside the cell, aiding in the study of biomolecular distribution in different cell compartments.

Influence of Nuclear Localization Sequences on the Intracellular Fate of Gold Nanoparticles

Daniela Drescher et al.

Summary: The study shows that the nuclear translocation of gold nanoparticles modified with nuclear localization sequences strongly depends on specific incubation conditions. Cells incubated with NLS-functionalized nanoparticles for 1 hour followed by a 24-hour chase time exhibit different intracellular distribution compared to continuous incubation for 24 hours. Enhanced uptake of NLS-functionalized nanoparticles compared to unmodified particles is observed under the same conditions. These results contribute to the optimization of nanoparticle analysis in various applications.

Summary: Gold nanostars are important tools in biophotonics and theranostics due to their ability to enhance local optical fields in living cells. SERS spectra show that the vibrational signals of molecules are similar in strength over a wide range of concentrations, making them ideal for label-free characterization of cells. Different intracellular processing of nanostars is influenced by their morphological properties, particularly the extent of hydrophobic interactions at the gold surface.
