The critical role of ligand topology: strikingly different properties of Gd(iii) complexes with regioisomeric AAZTA derivatives

Surprising Complexity of the [Gd(AAZTA)(H2O)2]- Chelate Revealed by NMR in the Frequency and Time Domains

Summary: Ln(III) complexes are usually isostructural, but the [Ln(AAZTA)(H2O)(x)](-) systems show structural variations causing changes in hydration number. NMR studies reveal different exchange dynamics processes of water molecules, with one Gd-bound water molecule exchanging 6 times faster than the other due to longer metal-water distance. Substitution of water molecule with fluoride in a diamagnetic-isostructural analogue allows for tracking chemical exchange process and describing thermodynamic behavior. NMR techniques provide a comprehensive understanding of structure and exchange dynamics of Ln-complexes.

Optimizing the relaxivity at high fields: systematic variation of the rotational dynamics in polynuclear Gd-complexes based on the AAZTA ligand

Summary: A series of polynuclear structures containing 2 to 6 GdAAZTA complexes were synthesized, with a significant increase in relaxivity observed from monomer to hexanuclear complex. The rotational dynamics of the multimers were analyzed through NMR study and DFT calculations, revealing effective relaxation enhancement strategies for different magnetic field strengths.MRI phantom images showed effective signal enhancement of the new MRI probes at clinically relevant magnetic field strengths compared to the clinically approved contrast agent ProHance (R).
