Engineered HaloTag variants for fluorescence lifetime multiplexing

Michelle S. Frei et al.

Summary: HaloTag variants offer unique brightness and fluorescence lifetimes compared to HaloTag7 when labeled with rhodamines, enabling multiplexed imaging and creating cell cycle indicators. These variants are powerful tools for fluorescence microscopy and can be used to generate fluorescence lifetime-based biosensors, with the brightest variant showing up to 40% higher brightness in live-cell imaging applications.

A General Strategy to Control Viscosity Sensitivity of Molecular Rotor-Based Fluorophores

Songtao Ye et al.

Summary: By chemically installing extended pi-rich alternating carbon-carbon linkages between the rotational electron donors and acceptors of RBFs, the lack of control of their viscosity sensitivity has been resolved. The length of the linkage strongly influences the viscosity sensitivity, providing the possibility to design RBFs with different viscosity sensitivities. RBFs can differentiate misfolded protein oligomers and insoluble aggregates in test tubes and live cells through a dual-color imaging strategy.

HaloTag Engineering for Enhanced Fluorogenicity and Kinetics with a Styrylpyridium Dye

Carla Miro-Vinyals et al.

Summary: This study reports the engineering of HaloTag for use with a chemically accessible and inexpensive fluorophore, achieving significant improvements in fluorogenicity and bioconjugation rate constants through directed evolution. The top variant, HT-SP5, enables reliable imaging in mammalian cells with a large fluorescence enhancement over the parent protein, highlighting the potential of engineering self-labeling proteins for alternative fluorophores.

Kinetic and Structural Characterization of the Self-Labeling Protein Tags HaloTag7, SNAP-tag, and CLIP-tag

Jonas Wilhelm et al.

Summary: The study compared the labeling kinetics and substrate specificities of self-labeling protein tags (SLPs) HaloTag7, SNAP-tag, and CLIP-tag. It was found that HaloTag7 reaches almost diffusion-limited labeling rate constants with some substrates, while SNAP-tag labeling rate constants are less affected by the structure of the label. Crystal structures of HaloTag7 and SNAP-tag labeled with fluorescent substrates helped explain their substrate preferences and aided in the design of substrates with improved labeling kinetics.
