Discovery of a potent SCAP degrader that ameliorates HFD-induced obesity, hyperlipidemia and insulin resistance via an autophagy-independent lysosomal pathway

Summary: A small molecule called lycorine was discovered to inhibit the SREBF pathway through a novel mechanism without inducing ER stress or activating NR1H3, improving dyslipidemia and fatty liver disease. Lycorine promotes SCAP lysosomal degradation through the SQSTM1-mediated autophagy-independent lysosomal degradation (SMAILD) pathway, leading to SCAP degradation without autophagy.
