Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Advances in Small-Cell Lung Cancer (SCLC) Translational Research

Benjamin J. Drapkin et al.

Summary: In recent years, there has been a renewed interest in the biology and therapeutic vulnerabilities of small-cell lung cancer (SCLC), leading to the development of diverse models and translational research avenues. These offer new hope for patients with SCLC by combining mechanism-based preclinical and clinical research.

An Epigenetic Mechanism Underlying Chromosome 17p Deletion-Driven Tumorigenesis

Mei Chen et al.

Summary: This study identifies PHF23 as a new tumor suppressor gene on chromosome 17p, which forms a previously unreported histone-modifying complex, the PSH complex, to regulate gene activation through a synergistic link between H3K4me3 and H3K27ac. Disruption of the PSH complex is crucial for the development and maintenance of PHF23-deficient and 17p-deleted tumors, revealing a novel epigenetic regulatory mechanism in 17p-deleted cancers.

Small-cell lung cancer

Charles M. Rudin et al.

Summary: Small-cell lung cancer is a type of lung cancer characterized by high proliferative rate, early metastasis, and poor prognosis, strongly associated with tobacco carcinogens. Most patients are diagnosed with metastatic disease, with only a small fraction having potentially curative earlier-stage disease. Genomic profiling reveals extensive chromosomal rearrangements and high mutation burden in SCLC, often involving inactivation of TP53 and RB1 tumor suppressor genes.

Roles of the HOX Proteins in Cancer Invasion and Metastasis

Ana Paco et al.

Summary: Invasion and metastasis are the leading causes of cancer-related deaths, with complex molecular networks and dependence on intra- and extracellular conditions and the premetastatic niche. Studies suggest an association between HOX gene expression levels and cancer cell invasion and metastasis, leading to the formation of new tumor masses. Deregulation of HOX genes by HMGA2/TET1 signaling and noncoding RNAs from the HOX loci can also promote invasion and metastasis by interfering with HOX gene expression or other genes relevant to these processes.
