Enzymatic C-to-C Protein Ligation

Summary: This study reports an engineered enzyme that accepts various nucleophile substrate mimetics, especially when a method to selectively inhibit the reactivity of byproducts is employed. In addition, C-terminal Leu-ethylenediamine motifs are identified as bona fide mimetics of native N-terminal sequences, enabling transpeptidase-catalyzed C-to-C ligations. This work significantly expands the synthetic scope of enzyme-catalyzed protein transpeptidation reactions.

Histone H2B Deacylation Selectivity: Exploring Chromatin's Dark Matter with an Engineered Sortase

Summary: We have developed a method to produce histone H2B through semisynthesis using an engineered sortase transpeptidase. Modified versions of histone H2B with specific acetylation, lactylation, and beta-hydroxybutyrylation at the N-terminal tail were incorporated into nucleosomes and studied as substrates for histone deacetylase (HDAC) complexes and sirtuins. Our findings indicate that these enzyme forms exhibit different rates and site-specificities, suggesting distinct biological roles in regulating chromatin structure and epigenetics.

Summary: Functionalized antibodies and antibody fragments play important roles in biomedical imaging, theranostics, and antibody-drug conjugates. This study describes a CRISPR/Cas9-based strategy to engineer hybridoma cells to secrete Fab' fragments bearing two distinct site-specific labeling motifs, enabling the generation of a stable cell line that secretes a dual tagged Fab' molecule. The technology platform developed in this study has the potential to advance the development of multimodal imaging agents, theranostics, and next-generation ADCs.

Laboratory evolution of a sortase enzyme that modifies amyloid-β protein

Summary: This study described the laboratory evolution of the bacterial transpeptidase sortase A to recognize specific sequences in the endogenous amyloid-beta (Aβ) protein, enabling site-specific protein modification without genetic manipulation. The evolved sortase showed a significantly changed substrate preference and was able to label endogenous Aβ in human cerebrospinal fluid with high sensitivity, demonstrating potential future applications in sortase-mediated labeling of Aβ peptides.
