The interplay of autophagy and oxidative stress in the pathogenesis and therapy of retinal degenerative diseases

Kun-Che Chang et al.

Summary: Oxidative stress caused by intracellular reactive oxygen species (ROS) production is closely associated with the pathogenesis of ocular diseases. Autophagy, a self-clearance pathway, plays an antioxidant role by regulating cellular ROS levels. However, excessive autophagy may lead to cell death. This review summarizes the mechanisms of interaction between ROS and autophagy and their roles in several ocular diseases.

Citicoline and Vitamin B12 Eye Drops in Type 1 Diabetes: Results of a 36-Month Pilot Study Evaluating Macular Electrophysiological Changes

Vincenzo Parisi et al.

Summary: The study demonstrated that patients with non-proliferative diabetic retinopathy (NPDR) who were treated with citicoline and vitamin B-12 eye drops for 36 months showed an improvement in macular function as detected by multifocal electroretinogram (mfERG) recordings. In contrast, patients with NPDR treated with a placebo during the same period exhibited a decline in macular function.

Fenofibrate increases circulating haematopoietic stem cells in people with diabetic retinopathy: a randomised, placebo-controlled trial

Benedetta Maria Bonora et al.

Summary: This study aimed to investigate the effect of fenofibrate on circulating haematopoietic stem/progenitor cells (HSPCs) in patients with diabetic retinopathy. The results showed that fenofibrate significantly increased the levels of HSPCs expressing CD34 and/or CD133, potentially explaining its role in reducing retinopathy progression.

Summary: High blood sugar leads to depletion of endogenous antioxidants in the retina, resulting in neurodegeneration and irreversible vision loss. Neurovascular Unit: A New Target for Treating Early Stages of Diabetic Retinopathy

Rafael Simo et al.

Summary: The concept of diabetic retinopathy has evolved to consider neurovascular unit impairment as a main target for therapeutic strategies. While neurodegeneration may not always be the primary event in the progression of the disease, phenotyping to identify patients who could benefit from neuroprotective treatments is recommended. Recent research has tested numerous neuroprotective treatments, including drugs with dual activity, aiming to address the complex nature of diabetic retinopathy.

Animal models of diabetic retinopathy

Jose Quiroz et al.

Summary: The retina is the layer of the eye that conducts light impulses to the optic nerve for vision, and diseases like diabetic retinopathy can lead to blindness. Diabetes affects millions globally and diabetic retinopathy is a common complication, highlighting the need for new therapeutics to address its underlying pathophysiology. Studying DR pathogenesis in animal models is crucial in developing treatments to repair damaged retinal tissue and reverse the disease.
