Multicomponent Solids of DL-2-Hydroxy-2-phenylacetic Acid and Pyridinecarboxamides

Summary: In this study, cocrystals of DL-2-Hydroxy-2-phenylacetic acid with pyridinecarboxamide isomers were prepared and characterized. The crystal structures showed the importance of hydrogen bonding and pi-pi stacking interactions in the stability of the cocrystals. The thermal stability and other physicochemical properties of the cocrystals were also evaluated and compared to the pure coformers.

Virtual Multicomponent Crystal Screening: Hydrogen Bonding Revisited

Summary: This research examines qualitative approaches based on hydrogen bonding strength. Molecular electrostatic potential (MEP) maps of 330 coformers were obtained from density functional theory simulations, and were used to inversely predict a number of APIs and their salts/cocrystals. Additionally, hydrogen bond energy (HBE) and hydrogen bond propensity (HBP) calculations were tested with various organic coformers, demonstrating the potential for a chemistry-based and ML combination to serve as a promising classifier for coformer selection.

The X-ray structure of a specific pyran-2,4-dione derivative was determined, revealing important O⋯H hydrogen bonds and closed-shell interactions. The compounds exhibit polar characteristics with different charged regions, as explored through molecular electrostatic potential maps.
