Higher yield and enhanced therapeutic effects of exosomes derived from MSCs in hydrogel-assisted 3D culture system for bone regeneration

Summary: In this study, a 3D culture model was engineered to enhance the osteoinductivity of MSC-derived exosomes. Exosomes derived from 3D culture showed higher yield and improved efficiency of collection compared to conventional monolayer culture. Moreover, these exosomes significantly promoted osteogenic gene and protein expression, proliferation, and migration of bone marrow mesenchymal stem cells. Mechanistic studies revealed that the upregulation of the YAP signaling pathway is the underlying mechanism. The 3D-Exos also resulted in enhanced new bone formation in rats with alveolar bone defects.

Exosomes derived from platelet-rich plasma administration in site mediate cartilage protection in subtalar osteoarthritis

Summary: This study aims to investigate the application of PRP-Exo in the treatment of subtalar osteoarthritis (STOA) caused by chronic ankle sprains. By incorporating PRP-Exo in a thermosensitive hydrogel (Gel), it was confirmed that Exo-Gel increased the retention of Exo in the joint and had a therapeutic effect. In vitro experiments showed that Exo-Gel promoted the proliferation and migration of bone mesenchymal stem cells and chondrocytes, as well as enhanced chondrogenic differentiation. In vivo experiments demonstrated that Exo-Gel inhibited apoptosis and hypertrophy of chondrocytes, enhanced proliferation, and potentially delayed the development of STOA.

Extracellular Vesicles from 3D Engineered Microtissues Harbor Disease-Related Cargo Absent in EVs from 2D Cultures

Summary: Engineered microtissues have a significant impact on extracellular vesicles (EVs) secreted by benign and malignant prostate cells, with a higher yield of EVs per cell in 3D cultures compared to 2D. Whole transcriptome sequencing and proteomics analysis reveal stark contrasts in molecular cargo between 2D-EVs and 3D-EVs, with exclusive enrichment of known drivers of prostate cancer progression in 3D-EVs. Functional assays demonstrate that only 3D-EVs confer increased invasiveness and chemoresistance to cells in 2D, highlighting the potential of engineered 3D microtissue cultures for identifying biomarkers not detectable in standard 2D conditions.
