Uptake, functionality, and re-release of extracellular vesicle-encapsulated cargo

This study demonstrates the functional delivery of EV-encapsulated RNA and protein cargo using luminescence and fluorescence reporters. mRNA-mediated delivery of gene editing tools to human primary muscle stem cells

Summary: In this study, we achieved efficient gene editing in human muscle stem cells using mRNA-based delivery of SpCas9 and a base editor. The edited cells retained their characteristics, proliferation capacity, and functional attributes. Furthermore, we demonstrated the highly efficient repair of a muscular dystrophy-causing mutation using mRNA-based delivery of a base editor. Therefore, this study establishes mRNA-mediated delivery of CRISPR/Cas9-based tools as a promising and universal approach for the clinical application of gene edited muscle stem cells to treat muscle disease.

Discovery and engineering of small SlugCas9 with broad targeting range and high specificity and activity

Summary: This study identified three compact Cas9 orthologs for mammalian genome editing, with SlugCas9 showing comparable activity to SaCas9 and recognizing a simple NNGG PAM. A SlugCas9-SaCas9 chimeric nuclease was generated with high specificity and activity, and a high-fidelity variant of SlugCas9 was engineered while maintaining high specificity without compromising on-target editing efficiency. These minimal Cas9 tools are ideal for both basic research and clinical applications.

Extracellular vesicles from dHL-60 cells as delivery vehicles for diverse therapeutics
