2-Diphenylphosphinonyl-acetyl as a Remote Directing Group for the Highly Stereoselective Synthesis of β-Glycosides

Xianglai Liu et al.

Summary: The utilization of the DPPA group in glycosylation allowed for efficient synthesis of challenging carbohydrates with excellent beta-selectivities and broad substrate scope. Additionally, the chemoselective removal of the DPPA group under mild Ni(OTf)(2) catalysis facilitated rapid conversion to biologically important molecules.

Halogen-bond-assisted radical activation of glycosyl donors enables mild and stereoconvergent 1,2-cis-glycosylation

Chen Zhang et al.

Summary: The chemistry of carbohydrates has a long history, and a simple, stereoselective and efficient glycosylation method is still needed for studying sugars. This study presents a strategy for 1,2-cis-glycosylation without using metals or strong acids. The method activates glycosyl donors through a radical cascade, resulting in the formation of 1,2-cis-glycosides with high stereoselectivity. The reaction shows remarkable tolerance to functional groups and is applicable to a broad range of donors.

Summary: This study reports the use of 2-(diphenylphosphinoyl)acetyl as a remote stereodirecting group in the direct synthesis of challenging beta-glycosides. The method showed high effectiveness in the synthesis of a deoxy-trisaccharide mimic of landomycin E's sugar chain and in the assembly of Citrobacter freundii O-antigens.

4CzIPN-tBu-Catalyzed Proton-Coupled Electron Transfer for Photosynthesis of Phosphorylated N-Heteroaromatics

Yan Liu et al.

Summary: 4,5,6-Tetrakis (3,6-di-tert-butyl-9H-carbazol-9-yl)-isophthalonitrile (4CzIPN-Bu-t) was developed as a photocatalyst for the phosphorus-radical-initiated cascade cyclization reaction of isocyanides. By using 4CzIPN-Bu-t as catalyst, a visible light-induced proton-coupled electron transfer strategy was developed for the generation of phosphorus-centered radicals, leading to the successful construction of phosphorylated phenanthridines, quinolines, and benzothiazoles.

Glycosyl-Radical-Based Synthesis of C-Alkyl Glycosides via Photomediated Defluorinative gem-Difluoroallylation

Cai-Yi Li et al.

Summary: A stereoselective, glycosyl radical-based method has been developed for the synthesis of C-alkyl glycosides via a photomediated defluorinative gem-difluoroallylation reaction. This method demonstrates the feasibility of glycosyl radicals participating in a photomediated radical polar crossover process, offering a diverse array of gemdifluoroalkene containing C-glycosides. Key features of this method include scalability, mild conditions, broad substrate scope, and suitability for late-stage modification of complex molecules.

1,2-trans-Stereoselective Synthesis of C-Glycosides of 2-Deoxy-2-amino-sugars Involving Glycosyl Radicals

Wen-Ze Shi et al.

Summary: The study reports for the first time that imidate radicals can efficiently add to glycals to produce glycosyl radicals, leading to a general, toxic-reagent-free synthesis of C-glycosides of 2-deoxy-2-amino sugars. The reaction is 1,2-trans-stereoselective and can utilize aryl alkenes as substrates. Late-stage functionalization and density functional theory calculations have been reported.

Nonenzymatic Stereoselective S-Glycosylation of Polypeptides and Proteins

Li-Qiang Wan et al.

Summary: This study presents a nonenzymatic glycosylation reaction that forms axial S-glycosidic bonds under biorelevant conditions. By using allyl glycosyl sulfones as precursors to glycosyl radicals and taking advantage of the exceptional functional group tolerance of radical processes, various unprotected glycosyl units can be selectively introduced to cysteine residues of peptides. The method was successfully applied for direct glycosylation of complex polypeptides and proteins after developing a second-generation protocol, with computational studies performed to elucidate the reaction mechanism.

Summary: C-Glycosyl peptides/proteins, stable mimics of native glycopeptides/proteins with therapeutic potential, have been successfully synthesized using a novel method. This synthesis approach is versatile, accommodating a wide range of coupling partners, including complex oligosaccharides and peptide substrates. The resulting vinyl C-glycosyl amino acids and peptides are amenable to further transformations for elongation of peptide and saccharide chains.

Radical philicity and its role in selective organic transformations

Faeze Parsaee et al.

Summary: Radical philicity, the ability of radicals to act as nucleophiles or electrophiles, is an important concept in organic chemistry. Traditionally, radicals are considered electron deficient, but the term "electron deficient" can imply electrophilicity in modern literature. This discrepancy has led to misconceptions about radical behavior, as many radicals actually exhibit nucleophilic behavior. This review aims to guide the understanding and prediction of radical philicity, demonstrating that not all radicals behave as electrophiles.
