Acellular biomaterial modulates myocardial inflammation and promotes endogenous mechanisms of postinfarct cardiac repair

Vishnu Vasanthan et al.

Summary: Through animal experiments, researchers found that implantation of porcine small intestinal submucosal extracellular matrix (SIS-ECM) can promote fibroblast-mediated angiogenesis and antifibrotic pathways, as well as reparative inflammatory response. This reparative inflammatory response may explain the previously reported functional improvements. The findings can provide insights for the design of next-generation cardiac repair materials.

Frontline Science: Conversion of neutrophils into atypical Ly6G+SiglecF+immune cells with neurosupportive potential in olfactory neuroepithelium

Kei Ogawa et al.

Summary: Recent research has shown that neutrophils can transform into double-positive cells expressing neurosupportive genes in the olfactory neuroepithelium (OE) under inflammatory conditions. These double-positive cells, derived from neutrophils, increase significantly during inflammation or tissue damage. Neutrophils change their surface markers and gene expression over time, expressing neurogenesis-related genes in addition to inflammation related genes.

Neutrophil Elastase Deficiency Ameliorates Myocardial Injury Post Myocardial Infarction in Mice

Yukino Ogura et al.

Summary: The study investigated the role of neutrophil elastase (NE) in the pathogenesis of myocardial infarction (MI) in mice. NE deficiency or inhibition improved survival and cardiac function post-MI, limited fibrosis, reduced heart-infiltrating neutrophils and inflammatory macrophages, and decreased apoptotic cells in the heart. NE deficiency might improve cardiac survival by upregulating insulin/Akt signaling, suggesting that inhibition of NE could be a novel therapeutic target in the treatment of MI.

Neutrophils Encompass a Regulatory Subset Suppressing T Cells in Apparently Healthy Cattle and Mice

Marion Rambault et al.

Summary: A new subset of regulatory neutrophils has been identified in mouse bone marrow and circulating in cattle blood, which can suppress T cell proliferation. These regulatory neutrophils, enriched in lymphoid organs, express distinct transcriptomic profiles and demonstrate contact-dependent suppression of T cell proliferation. These findings highlight the role of neutrophils as true partners of the adaptive immune response and suggest potential for new biomarkers and therapeutic interventions in controlling cattle diseases.

GPR34-mediated sensing of lysophosphatidylserine released by apoptotic neutrophils activates type 3 innate lymphoid cells to mediate tissue repair

Xiaqiong Wang et al.

Summary: Neutrophils play critical roles in tissue repair by stimulating the activation of ILC3s through the release of lysophosphatidylserine from apoptotic neutrophils, leading to enhanced IL-22 production and tissue repair. Deficiency of the GPR34 receptor in ILC3s limits IL-22 production and tissue repair in response to dying neutrophils, highlighting the importance of the interaction between neutrophils and ILC3s in tissue repair processes.

Skull and vertebral bone marrow are myeloid cell reservoirs for the meninges and CNS parenchyma

Andrea Cugurra et al.

Summary: The meninges contain a pool of monocytes and neutrophils supplied by adjacent skull and vertebral bone marrow, rather than by the blood. Under pathological conditions, CNS-infiltrating myeloid cells can originate from the brain borders and have distinct transcriptional signatures from their blood-derived counterparts. Therefore, myeloid cells from adjacent bone marrow niches are strategically placed to supply innate immune cells under both homeostatic and pathological conditions.
