Fisetin as a Senotherapeutic Agent: Biopharmaceutical Properties and Crosstalk between Cell Senescence and Neuroprotection

Osama Elsallabi et al.

Summary: As the average age increases in many Western countries, finding approaches to decrease the effects of aging on brain function becomes more urgent. Neuroinflammation and oxidative stress are two complex key factors in brain aging and chronic neurodegenerative diseases. Fisetin has been reported as a potent senotherapeutic that can extend lifespan by reducing peroxidation levels and enhancing antioxidant cell responses. It shows neuroprotective effects in various models of neurological disorders due to its actions on multiple pathways associated with different neurological disorders. This article aims to collect the recent achievements related to the antioxidant and neuroprotective effects of fisetin, as well as its pharmacokinetics, pharmacodynamics, and toxicity, and emerging drug delivery strategies to improve its bioavailability and chemical stability.

Acute-phase protein synthesis: a key feature of innate immune functions of the liver

Christian Ehlting et al.

Summary: The expression of acute-phase proteins plays a crucial role in maintaining homeostasis, tissue repair, and the organism's defense strategy, particularly in the context of innate immunity. Inflammatory responses lead to significant changes in serum protein composition, which can be used diagnostically. The liver, as the main site of APP synthesis, is constantly exposed to antigens or pathogens through blood flow, as well as systemic inflammatory signals.

Mannan-binding lectin deficiency augments hepatic endoplasmic reticulum stress through IP3R-controlled calcium release

Yu Chen et al.

Summary: MBL plays a critical role in ER calcium homeostasis and stress response, with its deficiency exacerbating liver injury susceptibility to ER stress. This study reveals a novel function of MBL in regulating ER stress and provides new insights into liver injury associated with MBL deficiency.

Molecular and cellular mechanisms of liver fibrosis and its regression

Tatiana Kisseleva et al.

Summary: Chronic liver injury results in liver inflammation and fibrosis, which can be reversed by eliminating activated myofibroblasts and resorbing the fibrous scar. Understanding the molecular mechanisms underlying liver fibrosis and its reversibility can lead to the identification of new therapeutic targets for liver fibrosis treatment.
