A One-Pot Synthesis of Glycans and Nucleosides Based on ortho-(1-Phenylvinyl)benzyl Glycosides

Rui Yang et al.

Summary: The study presents a one-pot glycosylation strategy for synthesizing both glycans and nucleosides based on ortho-(1-phenylvinyl)benzyl glycosides. It offers several advantages such as no aglycon transfers, no interference of departing species, and no unpleasant odor, and allows for the construction of up to four different glycosidic linkages.

Merging Reagent Modulation and Remote Anchimeric Assistance for Glycosylation: Highly Stereoselective Synthesis of α-Glycans up to a 30-mer

Yunqin Zhang et al.

Summary: The study presents an efficient strategy for synthesizing complex carbohydrates, successfully synthesizing branched Dendrobium Huoshanense glycans and linear Longan glycans. DFT calculations reveal the different selectivities of glycosylation reactions with various auxiliary groups. The orthogonal one-pot glycosylation strategy shows potential in the synthesis of complex glycans.

Discovery of Semi- and Fully-Synthetic Carbohydrate Vaccines Against Bacterial Infections Using a Medicinal Chemistry Approach Focus Review

Peter H. Seeberger

Summary: Researchers are using synthetic glycan-based medicinal chemistry to understand mammalian antiglycan immune response and define glycan epitopes for novel synthetic glycoconjugate vaccines against bacteria such as Streptococcus pneumoniae, Clostridium difficile, and Klebsiella pneumoniae. These efforts provide fundamental insights into how the human immune system recognizes carbohydrates and drive the discovery of carbohydrate vaccines.

An orthogonal and reactivity-based one-pot glycosylation strategy for both glycan and nucleoside synthesis: access to TMG-chitotriomycin, lipochitooligosaccharides and capuramycin

Haiqing He et al.

Summary: Glycans and nucleosides are vital in biological processes, and their chemical synthesis faces challenges such as stereochemical issues and poor solubility. A novel one-pot glycosylation strategy based on glycosyl ortho-(1-phenylvinyl)benzoates was developed, enabling efficient synthesis of both glycans and nucleosides with expanded scope and leaving group options. This strategy was successfully applied to the total synthesis of capuramycin and divergent synthesis of TMG-chitotriomycin, offering a straightforward route to these biologically important molecules.

Sequential activation of thioglycosides enables one-pot glycosylation

Lei Cai et al.

Summary: One-pot sequential glycosylations are straightforward strategies for constructing complex glycans, with sequential activation of thioglycosides being a powerful platform. This review summarizes the developments, recent advances, and synthetic utilizations of one-pot glycosylation based on relative reactivity value (RRV) controlled successive activation of thioglycosides, pre-activation based iterative activation of thioglycosides and the glycosyl sulfoxide activation initiated sequential activation of thioglycosides.
