Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Therapeutic potential of Fingolimod in triple negative breast cancer preclinical models

Tristan Rupp et al.

Summary: The study demonstrated that Fingolimod can inhibit tumor cell survival in TNBC and suppress tumor progression and metastases in preclinical mouse models without apparent adverse effects. This suggests that Fingolimod may have potential as a therapeutic option for TNBC and warrants further evaluation.

Prognostic Impact of Sphingosine Kinase 1 in Nonsmall Cell Lung Cancer

Nozomu Motono et al.

Summary: Bioactive sphingolipid pathway is relevant to lung physiology, and SPHK1 may be a prognostic factor and a potential drug target for lung-invasive adenocarcinoma. The study showed SPHK1 had positive correlation with fibroblasts and Ki-67 in both invasive adenocarcinoma, while having negative correlation with Ki-67 in squamous cell carcinoma.
