Recovery of particulate methane monooxygenase structure and activity in a lipid bilayer

Christopher W. Koo et al.

Summary: Bacterial methane oxidation using pMMO can be restored by reconstitution in nanodiscs, and the structures of pMMO in a lipid environment have been determined by cryo-electron microscopy. These findings provide important insights for understanding and engineering the function of pMMO.

A Pseudotetrahedral Terminal Oxoiron(IV) Complex: Mechanistic Promiscuity in C-H bond Oxidation Reactions

Katrin Warm et al.

Summary: In this study, a novel S=2 oxoiron(IV) species supported by a unique ligand was generated and characterized. This species exhibited high reactivity in hydrogen atom abstraction reactions and demonstrated a unique oxidative mechanism. Complex 2 can serve as an excellent electronic and functional model for biological oxoiron(IV) intermediates.

Wei Peng et al.

Summary: This study deciphers the catalytic cycle of pMMO in the presence of the physiological reductant duroquinol (DQH(2)), revealing that O-2 activation is initiated by the Cu-C(ii)-DQH(-) species. The research also uncovers the important roles of the phenol co-substrate for O-2 activation.

Coordination of the Copper Centers in Particulate Methane Monooxygenase: Comparison between Methanotrophs and Characterization of the CuC Site by EPR and ENDOR Spectroscopies

Richard J. Jodts et al.

Summary: This study found that the type II Methylocystis species strain Rockwell pMMO contains two monocopper sites similar to type I pMMOs, with the Cu-B site having an identical coordination environment. It also established, for the first time, the coordination environment of the spectroscopically observed Cu-C site in both types of pMMO, indicating its likely role in biological methane oxidation by pMMO.

Using X-ray free-electron lasers for spectroscopy of molecular catalysts and metalloenzymes

Uwe Bergmann et al.

Summary: X-ray lasers, with their tunable, intense and short pulses, offer unprecedented capabilities in studying molecular catalysts and metalloenzymes. The development of XFELs allows for powerful new probes of electronic structure dynamics, advancing our understanding of metalloenzymes. With more XFEL facilities starting to operate, new capabilities are expected in the future for real-time observation of metalloenzymes and catalysts in solution.
