The relationship between congenital heart disease and cancer in Swedish children: A population-based cohort study

Summary: This study found a connection between congenital heart disease (CHD) and childhood lymphomas and hepatoblastomas, especially in children with complex CHD. These results have the potential to increase awareness among physicians towards early cancer detection. Further research should focus on understanding the causal mechanisms underlying these findings, such as genetics and radiation exposure from cardiac procedures.

Association of Damaging Variants in Genes With Increased Cancer Risk Among Patients With Congenital Heart Disease

Summary: In this study, patients with congenital heart disease had a 1.3-fold increased frequency of damaging variants in cancer risk genes compared to control participants. The highest burden of damaging variants was found in cancer risk genes previously associated with CHD and those that regulate gene expression, especially in patients with extracardiac anomalies and/or neurodevelopmental delay. This suggests that genetic analyses of CHD patients could help identify causes of heart malformations and those at increased risk for cancer.

The Mutagenic Impact of Environmental Exposures in Human Cells and Cancer: Imprints Through Time

Summary: The accumulation of mutations in the genomes of cells is a result of continuous exposure to external damaging processes during life, leading to oncogenic mutations that are at the root of carcinogenesis. Whole genome sequencing data have revealed how cells gradually accumulate mutations due to exposure to various mutagenic processes. Dissecting mutation profiles based on altered bases has identified signatures reflecting past environmental mutagen exposures, ranging from chemotherapeutic drugs to genotoxic gut bacteria. Understanding how environmental factors induce mutations in healthy cells contributes to a better grasp of carcinogenesis and potential preventable causes of cancer.

Exposure to low-dose ionising radiation from cardiac catheterisation and risk of cancer: the COCCINELLE study cohort profile

Summary: The COCCINELLE study is a nationwide retropective French cohort aimed at evaluating the risk of cancer in patients who underwent cardiac catheterisation procedures during childhood. Findings to date show increased risk of cancer, leukemia, lymphoma, and solid tumors among the cohort compared to the general population. Future plans include dose reconstruction to estimate individual cumulative doses and conducting dose-response analysis.
