A composite System Combining Self-Targeted Carbon Dots and Thermosensitive Hydrogels for Challenging Ocular Drug Delivery

Summary: A composite system combining self-targeted carbon dots and thermosensitive in situ hydrogels was developed for ocular drug delivery. The system showed biphasic drug release and improved corneal transmittance and drug retention time. Cellular studies demonstrated good cytocompatibility and CD44 targeting of the system.

A study on poloxamer-based systems reveals that temperature-controlled events involve not only C-H stretching motions but also other vibrational regions, and optimal formulation results can be obtained through vibrational analysis at different temperatures.

Optimization of the Micellar-Based In Situ Gelling Systems Posaconazole with Quality by Design (QbD) Approach and Characterization by In Vitro Studies

Summary: In this study, PSC-loaded ocular micelles and in situ gels were developed and optimized for the first time. The optimized micelles showed high anti-fungal activity and no ocular toxicity, and remained in gel form even when diluted. These findings suggest that the optimized micellar-based in situ gel has the potential to enhance the ocular permeation and bioavailability of PSC.

Summary: In this study, prednisolone-containing ophthalmic formulations with cyclodextrin-based inclusion complex and a biopolymer were developed to enhance drug permeability and reduce toxicity. Results showed that compared to the conventional benzalkonium-chloride, the novel formulations exhibited lower cytotoxicity and higher drug permeability. These findings suggest the innovative potential and promising application of the new non-toxic ophthalmic formulations.

The optimized formulation (MEL-HSA-P14%) showed enhanced permeability through the blood-brain barrier-specific lipid fraction and promise for overcoming challenges of nasal drug delivery.

Summary: This study focused on incorporating itraconazole into nanocrystalline carrier system with Pluronic (R) F127 stabilizer and formulating it into thermosensitive in situ ocular gel, showing improved antifungal activity against Candida albicans. The optimized gel base demonstrated better therapeutic effect on infected porcine eye models, with a 93% decrease in the CA population after 48 hours.
