In Vitro Biosynthesis of the [Fe]-Hydrogenase Cofactor Verifies the Proposed Biosynthetic Precursors

Sebastian Schaupp et al.

Summary: In this study, an in vitro biosynthesis assay of the FeGP cofactor was conducted using cell extract of a specific bacterium, revealing that pyridinol derivatives 1, 2, and 3 are precursors of the FeGP cofactor, with the carboxy group of 3 being converted to the acyl ligand.

On the Evolutionary History of the Twenty Encoded Amino Acids

Andreas Kirschning

Summary: This report focuses on the prebiotic and metabolic aspects of amino acids and proteins, as well as the major biosynthetic pathways. Coenzymes play a key role in amino acid metabolism, being linked to glycolysis and the tricarboxylic acid cycle. Thus, the report provides additional insights into the molecular evolution of primary metabolism.

Mechanistic concepts of iron-sulfur protein biogenesis in Biology

Joseph J. Braymer et al.

Summary: Iron-sulfur (Fe/S) proteins play essential roles in various cellular processes and their synthesis requires assistance from specific protein machineries. These systems differ between prokaryotes and eukaryotes, underlining the complexity and specificity of Fe/S protein biogenesis.

Four families of folate-independent methionine synthases

Morgan N. Price et al.

Summary: The study investigated the roles and mechanisms of different families of methionine synthases in methionine synthesis, identifying four folate-independent methionine synthase families. Some of these families co-occur with corrinoid proteins, while another family interacts with oxygen for activity.

Structure and function of aerotolerant, multiple-turnover THI4 thiazole synthases

Jaya Joshi et al.

Summary: Plant and fungal THI4 thiazole synthases produce thiamin thiazole in aerobic conditions via a single-turnover suicide reaction, while multiple-turnover THI4s can operate in aerobic, mild, low-sulfide conditions. These THI4s lacking active-site Cys have been found in both anaerobic archaeal methanogens and aerobic mesophiles, indicating a wider potential range of operation for THI4 enzymes.
