Cardiotoxicity of cyclophosphamide's metabolites: an in vitro metabolomics approach in AC16 human cardiomyocytes

Summary: This study investigates the cardiotoxic mechanisms of cyclophosphamide and its main biotransformation products through classic and metabolomics studies. The results show that 4-hydroxycyclophosphamide and acrolein cause significant cardiotoxicity at low concentrations, while the toxicity of cyclophosphamide is higher. Metabolomics studies reveal metabolic changes in the heart and impairment of precursor amino acids used in glutathione synthesis after exposure to cyclophosphamide's metabolites.

Recent studies suggest that heart failure may promote cancer progression, but whether cardiac remodeling without cardiac contractile dysfunction can promote cancer progression remains uncertain. In this study, using a mouse model, it was found that low-dose PE infusion can promote tumor growth and is associated with abundant secretion of cardiac and tumor factors. Therefore, early detection and treatment of hypertension may benefit the prognosis of cancer patients.

Cardiotoxicity evaluation and comparison of diterpene alkaloids on zebrafish
