Kidney transplantation from triple-knockout pigs expressing multiple human proteins in cynomolgus macaques

Summary: The study demonstrates that long-term survival of renal xenografts from TKO pigs can be achieved in cynomolgus monkeys through the expression of additional human transgenes. The level of CRP expression was found to be associated with recipient survival time.

Dietary Neu5Ac Intervention Protects Against Atherosclerosis Associated With Human-Like Neu5Gc Loss-Brief Report

Kunio Kawanishi et al.

Summary: Species-specific pseudogenization of the CMAH gene during human evolution eliminated Neu5Gc biosynthesis from its precursor Neu5Ac. Interventional Neu5Ac feeding can mitigate or prevent the increased risk for atherosclerosis mediated by red meat/Neu5Gc, and has an intrinsic protective effect.

Histopathology of pig kidney grafts with/without expression of the carbohydrate Neu5Gc in immunosuppressed baboons

Summary: The study found that pig kidney grafts lacking Neu5Gc expression were associated with increased binding of baboon antibodies to pig endothelium and reduced graft survival when transplanted into baboons.

The role of neuraminidase 1 (NEU1) in cytokine release by primary mouse mesangial cells and disease outcomes in murine lupus nephritis

Jessalyn Rodgers et al.

Summary: Reducing NEU1 expression may reduce inflammatory response in lupus nephritis. In experiments inducing kidney disease in lupus-prone mice, NEU1 appears to mainly mediate cytokine release by mesangial cells. However, the impact of reduced NEU1 levels on disease may be minor in lupus-prone mouse colony.

Small RNAs are modified with N-glycans and displayed on the surface of living cells

Ryan A. Flynn et al.

Summary: This study challenges the conventional view that RNA is not a major target of glycosylation by providing evidence that mammals use RNA as a third scaffold for glycosylation. The researchers found that conserved small noncoding RNAs bear sialylated glycans, which are present in multiple cell types and mammalian species. The majority of glycoRNAs were located on the cell surface and were shown to interact with anti-dsRNA antibodies and members of the Siglec receptor family, suggesting a direct interface between RNA biology and glycobiology.

Summary: Genetic modification of Yucatan miniature pigs for xenotransplantation should be specific to the target organ, and silencing additional genes such as CMAH or A3GALT2 based on GTKO may not be essential.

You cannot stay in the laboratory forever: Taking pig kidney xenotransplantation from the laboratory to the clinic

David K. C. Cooper et al.

Summary: Collaboration between the laboratory team and the clinical team is essential to prepare for the first clinical trial of life-supporting kidney transplantation in the genetically-engineered pig-to-nonhuman primate model. Determining optimal genetic modifications, immunosuppressive therapeutic regimens, and measures to minimize infectious risks are crucial for the progress of xenotransplantation.
