Polyhydroxylated bergamotane-type sesquiterpenoids from cultures of Paraconiothyrium sporulosum YK-03 and their absolute configurations

Summary: Eight undescribed polyhydroxylated bergamotane-type sesquiterpenoids were discovered from a marine mud-associated fungus, and their chemical structures and absolute configurations were successfully determined. Among them, sporuloketals A-B represent two rare tetracyclic compounds. This study also validated the application of ECD empirical rules for determining the absolute configurations of these bergamotane-type sesquiterpenoids for the first time.

Asymmetric Total Synthesis of (-)-Phaeocaulisin A

Summary: The therapeutic properties of Curcuma have been long known, and recent studies have focused on the biological activities of guaiane-type sesquiterpenes extracted from it. Among these compounds, phaeocaulisin A has shown great potential as an anti-inflammatory and anticancer agent. The researchers have successfully implemented the first enantioselective total synthesis of phaeocaulisin A, which provides a synthetic platform for further studies and drug design.

Guided isolation of daphnane-type diterpenes from Daphne genkwa by molecular network strategies

Summary: A molecular networking-guided study successfully isolated twelve daphnane-type diterpenoids, including four undescribed compounds, from Daphne genkwa Sieb. et Zucc. The structures of these compounds were determined through spectroscopic analyses, ECD calculations, and single-crystal X-ray diffraction analysis. The isolates demonstrated inhibitory activity against A549 cells, with IC50 values ranging from 7.77 to 20.56 μM, and a preliminary discussion on their structure-activity relationship was conducted. Further experiments revealed that five of these compounds appeared to inhibit A549 cell lines by inducing apoptosis.
