SynPathy: Predicting Drug Synergy through Drug-Associated Pathways Using Deep Learning

Yi-Ching Tang et al.

Summary: This study proposes a biologically-motivated deep learning approach to predict drug synergy and quantify interactions in drug combinations using molecular data. The method outperformed previous approaches and provided potential candidate pathways to explain the predictions. The study also suggests that drug combinations tend to be more synergistic when their contributing pathways are closer on a protein interaction network, offering a potential strategy for combination therapy.

IL-12 Family Cytokines in Cancer and Immunotherapy

Bhalchandra Mirlekar et al.

Summary: The IL-12 family cytokines are crucial in regulating tumor immune contexture and activating anti-tumor immune responses. Different family members have distinct roles, encompassing both promoting effector responses and inhibitory functions. These cytokines hold promise as candidates for developing immunomodulatory therapeutic approaches.

Facilitating Drug Discovery in Breast Cancer by Virtually Screening Patients Using In Vitro Drug Response Modeling

Robert F. Gruener et al.

Summary: Integration of in vitro drug screening data and patient tumor molecular information led to the creation of a virtual drug screening pipeline enabling drug discovery with simultaneous biomarker identification for a patient population. Using this pipeline, the study successfully identified a potential compound for triple-negative breast cancer and validated its efficacy in both in vitro and in vivo settings.

Tumour-targeted interleukin-12 and entinostat combination therapy improves cancer survival by reprogramming the tumour immune cell landscape

Kristin C. Hicks et al.

Summary: Combining the histone deacetylase inhibitor, Entinostat, with the recombinant murine immune-cytokine NHS-rmIL12 can reprogram the tumor microenvironment and lead to tumor eradication. This combination therapy promotes the action of functional CD8(+) T cells and activated neutrophils, driving macrophage polarization and benefiting overall survival in multiple human tumor types.
