Synthesis, molecular docking and molecular dynamics studies of novel tacrine-carbamate derivatives as potent cholinesterase inhibitors

Summary: In this study, new tacrine derivatives containing carbamate group were synthesized and evaluated for their inhibition activities against AChE and BuChE. The synthesized compounds showed strong inhibitory effects on both cholinesterases, with 6k exhibiting the best inhibitor activity and 6b showing the highest selectivity. Molecular modeling approaches were used to understand the interaction between the compounds and proteins, and covalent docking approaches were applied to determine the binding modes of the novel compounds at the binding sites of cholinesterase enzymes.
